The invention describes an improved and advantageous process of preparation of 4-(4-butyl amino)-2,3- tetramethylenequinoline hydrochloride (Centbucridine), 4-substituted amino-2,3-polymethylenequinoline hydrochlorides, and similar derivatives thereof, useful local anaesthetic, in treatment of neurodegenerative disorders, stress, stroke, dementia, antimalarial, sexually transmitted diseases, anti-amoebic, bactericidal, anti-protozoal, anti-cancer, immune diseases, cardiovascular diseases, diabetes, obesity, inflammation. This process reduces number of synthetic steps for synthesis of these compounds in excellent yield and high purity and avoids hazardous conditions like use of phenol solvent, strong base i.e. NaH and reactions in sealed bomb at elevated temperature.
