Project Overview

In the proposed study based on in silico gene network analysis, associated secondary resistance genes will be studied for their transcriptional expression using antibiotic as signal molecule. Laboratory strain, clinical isolates, mutants and clones will be used in the study. mRNA sequence of secondary resistance genes will be done to understand the sequence variation of over expressed one from normal one. A quantitative PCR based assay will be developed to detect multidrug resistance using secondary resistome as biomarker which will be simple and a reliable tool in diagnostic microbiology laboratory.

Achievements

Characterization of 500 isolates were completed. Transcriptional expression analysis of mrcA, mrcB and mtgA was performed by Real- Time PCR S.N. Cloning of full-length OXA-10, OXA-23 and VEB1 genes in araBAD-induced pBAD-18 CAM vector is done. OXA-20 and VEB-1 were constructed by gene synthesis. Antibiotic susceptibility assay and Biofilm formation upon in vivo expression of OXA- 10 and OXA-23 beta-lactamase are done.Cloning, Expression, Purification, and Enzyme kinetic analysis of OXA-10, OXA- 20 and OXA-23 beta-lactamase is performed.
