Project Overview
Transition metal-catalyzed reactions have become standard practices for forming C–C and C–X bonds, paving the way for innovative approaches in organic synthesis. In this regard, the design of metal-carbene systems that can provide structural diversity in fewer synthetic steps is a key objective for achieving both economic and environmental goals. This research proposal focuses on establishing effective and advanced methods for transition-metal-catalyzed carbene-mediated reactions through processes such as annulation, insertion, rearrangement, and cycloaddition to produce structurally varied heterocycle scaffolds with potential biological significance. We will evaluate commercially available catalysts to identify the most effective catalytic system, and we will develop strategies that utilize commercially obtainable starting materials or enable the straightforward preparation of substrates from inexpensive fine chemicals to ensure the success of this project. Isotopic labeling experiments and spectroscopic techniques, including NMR and ESI/LC MS, will be carried out to elucidate the mechanisms of each developed methodology. Additionally, efforts will be made to isolate and characterize key intermediates, including single-crystal X-ray analysis. The established methodologies will also be applied to the potential total synthesis of natural products featuring targeted scaffolds, along with further modifications aimed at enhancing their biological efficacy.
