Project Overview

Intrauterine adhesions or fibrosis develop due to acquired damage in the endometrium, resulting in partial to complete dysfunction of the endometrium within the uterine cavity. These intrauterine adhesions, with symptoms including infertility, amenorrhea, and hypomenorrhea, are known as Asherman’s syndrome (AS). The present treatment methods are ineffective, resulting in recurrent fibrosis. This proposal aims to develop a biomimetic estrogen receptor targeted gelatin methacrylate (GelMA) system to deliver perfinidone for targeting the treatment of Asherman syndrome and determining the therapeutic efficiency in preventing in vitro and in vivo. Targeting the estrogen receptor is achieved by conjugating estradiol to the hydrogel. The combination of pirfenidone and D-alpha-tocopheryl polyethylene glycol 1000 succinate (TPGS) helps in fibrolysis through their respective tissue plasminogen activator and antifibrolytic properties. The targeted synergistic effect with our drug delivery system could be a powerful tool to study other gynecological disorders. This research introduces a targeted drug delivery system for Asherman’s syndrome, promising advancements in treatment by specifically targeting the dysfunctional endometrium with a novel GelMA hydrogel loaded with pirfenidone (P) and TPGS.
