About
The study aims to investigate the molecular mechanism of cytoplasmic DNA degradation by TREX1, a nuclease involved in normal cell DNA regulation. The research suggests that cytosolic DNA might be endogenously alkylated and resist TREX1-mediated degradation, leading to the accumulation of alkylated DNA in the cytosol. The project will use high-resolution mass-spectrometry to examine endogenous alkylation in cytoplasmic DNA, synthetic oligonucleotides containing specific DNA alkyl-adducts, and in vitro assays to determine the effect of DNA containing abasic sites or methyl-adducts on CGAS activity.
