Project Overview

Lipin-1 is a crucial protein that plays a crucial role in maintaining cellular metabolic homeostasis and controlling cellular growth and proliferation. Its dysregulation has been linked to the onset and development of cancer, with its expression levels correlated with tumor aggressivity and poor prognosis. The pathways determining lipin-1's role in cancer are complex and multifactorial. studies have shown that lipin-1 has a role in cell cycle progression, and its inhibition or knockdown causes spindle abnormalities. Additionally, lipin-1 modulates the Akt1 signaling pathway, a cellular serine or threonine protein kinase involved in cell cycle regulation and key cellular functions. Akt1 has been implicated in cancer development and progression, and targeting it has emerged as a promising approach for cancer therapy. Lipin-1 is a downstream target of the PI3K-Akt-mTOR pathway and a bona fide target of mTORC1 kinase that phosphorylates it. The study aims to understand the precise mechanism behind the relationship between lipin-1 and Akt1 or mTORC2 and study its significance in mammalian cells, particularly in cancers. Lipin-1 is an intriguing protein that needs further investigation in cancers and metabolic disorders due to its known role in lipid metabolism regulation and the proposed mechanism of lipin-1 in cell cycle progression and PI3K-Akt-mTOR signaling.
