The project aims to develop low-cost, field-deployable sensors for detecting pathogenic bacteria, such as Escherichia coli, which cause severe diarrhea and death. India's 'Jal Jeevan Mission' aims to accelerate the focus on safe water supplies. Nano-biosensors incorporating analyte-specific biomimetic receptors and nanomaterials with exceptional optoelectronic properties are being explored for their ability to detect pathogens. Novel nanomaterials like MXenes and fluorescent nanostructures (FNs) have gained attention for their sensitive fluorescent detection of pathogens. These materials have the ability to adsorb ssDNA, increase target-induced ssDNA release, and serve as quenchers. Highly specific biomimetic receptors like Aptamer, DNAzyme, and CRIsPR or Cas can enhance the sensitivity and flexibility of these sensors. The project proposal aims to synthesize MXenes and FNs to exhibit enhanced FRET pairing, facilitating sensitive and specific detection of REP strains like E.coli. The integrated sensors concept will boost point-of-care device accuracy, selectivity, and detection limit for pathogenic bacteria. Additionally, a disposable electrodes-based fluorescence nanoprobe will be developed for easy and cost-effective on-site detection of pathogens.
